RC-1291

RC-1291 is Sapphire Therapeutics’ lead ghrelin mimetic with first-in-class potential in the treatment of anorexia and cachexia in cancer patients. Although anorexia and cachexia frequently accompany malignant disease states, there are currently no effective treatments approved for this indication.

RC-1291 is a synthetic, small molecule ghrelin mimetic that is orally available and intended to be administered as a convenient once-daily treatment to cancer patients suffering from anorexia and cachexia. Intervention with RC-1291 is expected to improve appetite and food intake and attenuate or reverse ongoing weight loss and loss of lean body mass. Additional benefits may include improvements in functional performance and quality of life.

RC-1291 has received Fast Track designation from the FDA for anorexia and cachexia in cancer patients.

Ipamorelin

Ipamorelin may potentially address several gastric motility disorders. The first indication for which ipamorelin is being developed is post-operative ileus (POI). POI is the slowing or stopping of intestinal motility that occurs following surgery.

POI is associated with complications such as gastrointestinal perforation, hospital acquired infections, malnourishment, and muscular atrophy. In addition, delayed gastrointestinal recovery contributes to prolonged length of hospital stay and a reduced quality of life due to discomfort, anxiety, additional pain, and the inability to eat liquid or solid foods.

Currently available options for the management of POI (nothing by mouth, IV fluids, nasogastric suction, dopamine antagonists, laxatives, etc.) are generally considered ineffective.

Ipamorelin is a synthetic, small molecule ghrelin mimetic intended to be administered by IV infusion. Ipamorelin offers an opportunity to advance the standard of care for the management of POI, potentially resulting in better management of the patient. The compound has undergone Phase I testing in normal human volunteers and will be advanced into studies in surgical patients.

RC-1141

RC-1141 is a synthetic, orally available, small molecule ghrelin mimetic that is at the pre-IND stage of development for opioid-induced bowel dysfunction.
